Dysfunction of desmosomes is associated with the pathogenesis of cutaneous diseases. We identified streptococcal pyrogenic exotoxin B (SpeB) as a proteolytic factor that cleaves the extracellular domains of desmoglein 1 and 3. 2001-11-01 SpeB has been shown to increase internalization (Tsai et al., 1998), two other studies have demonstrated decreased internalization with SpeB-producing bacteria (Burns et al., 1998;Chausseeetal.,2000).Finally,onestudyindicatesthat the effect of SpeB on internalization is dependent upon the amount of hyaluronic acid capsule present in the strain 2017-10-03 speB and spi are contiguous in the streptococcal genome. Activity of SpeB was determined by active site titration using the specific cysteine proteinase inhibitor L-trans-epoxysuccinylleucylamido(4-guanidino)butane (E-64) as previously described . We also show that EndoS only hydrolyzes the glycan moiety on native but not denatured IgG. Thus, SpeB has a broad immunoglobulin-degrading activity, while EndoS is highly specific for IgG.
